What STEP 9 measured
The 2024 STEP 9 trial randomized 407 adults with obesity and painful knee osteoarthritis to semaglutide or placebo, alongside lifestyle counseling, for 68 weeks. Women made up 81.6% of participants. Average pain scores fell by 41.7 points with semaglutide and 27.5 with placebo; average weight fell by 13.7% and 3.2%, respectively.
Keep the question precise
These results concern pain and weight in a defined population. They do not demonstrate regenerated cartilage or prove that women respond differently from men. S7’s practical reading question is whether a headline names the outcome actually measured. A pain questionnaire and an image of joint structure would address different claims.
Benefits and limits belong together
Adverse events led to permanent discontinuation in 6.7% versus 3.0%; gastrointestinal problems were the most common reason. Novo Nordisk funded the trial.
